The guy says perform 300 reps for each exercise but it's actually only 30. 300 would be impressive but this routine isn't. He's using pretty light weights, I'd say anyone with halfway decent endurance should be able to knock this out without a lot of trouble. This guy's physique is probably also due to a lot of other workouts, good dieting, and genetics as much as it is to this routine.
The trainer for the movie actually had his actors doing very different workouts each time so there is no standard 300 workout but there are a few of the routines he created available on the internet. I don't think this is an actual one, just one this guy made up. There are a lot more impressive workouts on your link like this one. This is an actual one from the movie trainer:
25 pull-ups
50 - 135 (60kgs) deadlifts
50 pushups
50 24" box jumps
50 floor cleaners
50 dumbbell or kettlebell clean and presses with 36lbs (25 each side)
25 pull-ups
The key is to finish in 20 minutes. Again, it's all about endurance. I did it with a friend of mine and didn't find it all that challenging so I subbed in 5x5 benchpress with 225lbs instead of the pushups which stretched the workout to 30 minutes but I still felt pretty good after so I hit the heavy bag for 15 min.
Here's my breakdown on the time to get it under 20 minutes.
25 pull-ups 1 min
50 deadlifts 2 min
50 pushups 1 min
50 box jumps 1 1/2 min
50 floor cleaners 5 min
50 dumbbell or kettlebell clean and presses with 36lbs (25 each side) 1 1/2 min
25 pull-ups 3 min
You're not supposed to rest between sets but I think we can reasonably assume there will still be about 5 min down time as you switch from station to station. This brings us to 20 min.
I think this would be hard if you were one of the actors on the movie since you're constantly under the strain and stress of other workouts and extreme dieting to get cut which doesn't leave you any reserves. But if you're a fit guy with 10% body fat or more you should actually have an easier time of it.

btw i just saw this movie for the first time last night. all the cast were in pretty good shape, but OMG what a horrible fkn movie!!

It's about Leonidas and his Spartans yelling and being awesome and killing a bunch of Persians. Don't think too hard about it.

The Gorgo subplot was horrible, tho.
